Abstract

In Planta Transformation of SoSPS1 Gene Through Agrobacterium tumefaciens in Balinese Grapevine (Vitis Vinifera L. var. Alphonso Lavalle). Vitis vinifera L. var. Alphonso Lavalle is one of the cultivar of grapevine cultivated in Bali. The sour taste of fruit is one of the problem of these cultivar that make it is less prefer by consumer. Research regarding improvement of fruit quality through genetic engineering has been investigated. The aims of the research were to find out the most proper methods of in planta transformation mediated with Agrobacterium tumefaciens applied on V. vinifera. The T-DNA harbored the SoSPS1 gene under the control of promoter of the CaMV from Cauliflower Mosaic Virus and contained the NPTII gene, a kanamycin resistant gene as a selectable marker for transformant selection. V. vinifera seedlings from stem cutting aged 40-50 days were ripped and then dipped with the bacterial suspension for 1 hour, 2 hours, 1 day, and 2 days. Visualization of V. vinifera samples after PCR showed that ripping and dipping for 2 hours method was positively integrated with NPTII gene. The dipping of plants wound for 2 hours treatment was effective for inserting gene of interest in ripping and dipping methods.

Abstract

Effectiveness Test of Several Concentrations of Kirinyuh Leaf Extract (Chromolaena odorata L.) Toward Development of Cabbage Crop Caterpillar (Crocidolomia pavonana F.). This research was conducted at the Laboratory of Plant Pests and Diseases of the Faculty of Agriculture, Udayana University from April 2018 to August 2018 to test the extract of kirinyuh leaves (Chromolaena odorata) on the development of biology of cabbage crop caterpillar (Crocidolomia pavonana). The completely randomized design (CRD) was used in this study, with four types of treatment of leaf extract concentration of 10%, 20%, 30%, 40%, and control. Each treatment was repeated 10 times. Observations were completed every day of the larval stage until the larvae do not carry out any activity anymore (dead), or until they become imago. The results showed that (1) the leaf extract of C. odorata plant which was used as research material had the potential to suppress the biological development of C. pavonanadi larvae in the laboratory, (2) the application of leaf extract concentration of 10% and 20% had an influence on the development of larvae which had developed slow compared to concentrations of 30% and 40%, where larvae experience rapid development of only 9 observations and (3) the most effective test leaf extract treatment was the treatment of leaf extracts of C. odorata concentrations of 40% and 30% followed by leaf extract concentrations of 20% and 10%.

Abstract

Isolation and Identification of Endomycorrhizal Indigenous on Salak Rhizosphere in Karangasem Regency and its Propagation. This research aimed to determine and obtain AMF indigenus isolates and their propagation at different levels of soil water content. This research used Completely Randomized Design with 2 Factors. The first factor was location of soil sampling, consisting of 3 levels of treatment (Location 1, Location 2, and Location 3) and the second factor was soil water content consisting of 3 levels of treatment (100, 70, and 40% field capacity). The results of isolation and identification indicate that there were two genus of AMF found on salak rizosphere in Karangasem Regency, those were Glomus and Gigaspora with several different species in it. The percentage of root infection rate was 100% and was categorized very high. The result of AMF spore propagation showed that combination treatment of location 1 which sandy clay soil textured and 40% field capacity produced the highest spores i.e 2382 spores or rised 2282% from 100 spores which were inoculated.

Abstract

Identification and Characterization of Plant Genetic Resources Local Fruits in Jembrana. Fruits are one source of nutritious food that have diverse functions such as: source of vitamins, minerals, and fiber needed in the daily menu. Fruits are used as community consumption, tourism needs, inter island trade, as well as for religious activities, even thoughthere are local fruit are used in “usadha”by generations. Jembrana Regency has various types of fruits. This study aims to identify the genetic resources of fruits in Jembrana Regency. This research was conducted in five stages: (1) collected secondary data, (2) surveyed the types of genetic resources and their distributions, (3) identification of morphological and agronomiccharacters, (4) determined superior fruit, and (5) compiled geographic map the distribution of genetic resources of local fruits. The result of research found 38 species and 53 sub-types of local fruit plants, with 6 types of superior fruit, 12 type of rare fruit, 17 types of fruit for upakara, and 9 types of fruits for medicine. Based on the survey conducted in Jembrana District, 41 geographical distribution maps of local fruits can be compiled.

Abstract

The Effectiveness Test of Indigenous Endomycorrhiza on Maize Plant Growth (Zea mays L.). Efforts to increase crop productivity were hampered due to various obstacles such as genetic, environmental, and biotic factors, in order that alternative technologies such as endomycorrhizal biological agents were required. This study was aimed to determine the effectiveness of indigenous endomycorrhiza for the growth of maize. The experiments were carried out in the laboratory and greenhouse. The field experiment was designed in a nested randomized block design. The first factor was genus of indigenous endomycorrhiza (M) consisting of 3 lavels i.e genus Glomus (G), genus Gigaspora (F) and the mixed genus (C), the second factor was the spore dose (D) consisting of 3 levels i.e. D1 = dose of 50 spores, D2 = 100 spores and D3 = 150 spores. The genus Glomus was able to increase the weight of fresh and dry oven total stalk, the number of leaves and the height of the plants and those significantly higher than that of the genus of Gigaspora and the mixed genus. Spore doses (50, 100 and 150 spores) of the genus Glomus and the genus Gigaspora and mixture gave no significant different results on all observed variables.

Abstract

Damage and Product Losses by Fruit Flies Bactrocera dorsalis Complex (Diptera: Tephritidae) at Citrus Cultivation. This research knows the damage and nominal loss caused by fruit flies attack on citrus cultivation. The study took place from May to September 2016, from fruit to harvest, fruit fly observations were performed using relative methods. The results showed the damage of fruit at citrus cultivation in place of research have category relative moderate until heavy. Of the 10 plant samples observed 3 damaged plants reached 51%. Then 7 other plant samples suffered damage in the medium category. But the average damage reached 44%, this is a fruit fly is categorized as a very harmful pest. Nominal losses in 10 sample plants observed with the number of fruit fell as much as 943 fruit and fruit weight reached 157.17 kg obtained nominal loss of Rp. 471,500. Based on the data obtained hectar loss of Rp.29.468.750. Fruit fly attacks have increased along with the condition of citrus fruits that begin to experience physiological development so that the loss of the number of fruit fall that resulted in economic losses.

Abstract

Flight Activities and Population Development of Thrips parvispinus Karny (Thysanoptera: Thripidae) on Chili (Capsicum annuum L.). Thrips parvispinus Karny (Thysanoptera: Thripidae) is one of the main pests on chili. T. parvispinus attacked the leaves and flowers on chili. The behavior of its attacked fluctuate between morning until late afternoon since the plants grow in the field. The purpose of this research was to know the flight activity of T. parvispinus and its population development on leaf and flower of chilli plant. The research used survey method with direct observation on plants in the field. Flight activity of T. parvispinus was measured by using a yellow sticky trap placed diagonally on the plant. The results showed that daily flight activity of adult T. parvispinus fluctuated between morning, afternoon, and evening. The highest flight activity of adult occurred at 09.00-10.00 (39.81 adults) and the lowest was at 18.00-19.00 (9.84 adults). The population of T. parvispinus nymph was more than adult in the leaf. The highest population of nymphs occurred at 12.00-13.00 (21.07 nymphs). While the population of T. parvispinus adul was more on flower than nymph population. The highest adult population occurred at 18.00-19.00 (27.36 adults). There were strong indications that the nymph stadium frevers live on the part of the leaf than the flower, while the adult prefers the flower rather than the leaf.
